The G-protein-coupled receptor (GPCR) autoproteolysis-inducing (GAIN) domain, includes GPCRs involved in adhesion with a characteristic autoproteolysis motif of HLT/S known as the GPCR proteolysis site (GPS). GPS is also shared by polycystic kidney disease (PKD) proteins. GAIN has been shown to be both necessary and sufficient for autoproteolysis [1].
